Abstract:
Tizanidine is a centrally acting α2-adrenergic agonist widely used as a muscle relaxant in the management of spasticity and musculoskeletal disorders. However, its conventional oral dosage forms suffer from limitations such as extensive first-pass metabolism, low bioavailability, and delayed onset of action. The present review focuses on the development and evaluation of hydrogel-based mouth dissolving films (MDFs) of tizanidine as an innovative drug delivery system. Hydrogel-based MDFs combine the advantages of hydrogels, such as high water absorption and flexibility, with the benefits of oral films, including rapid disintegration and ease of administration. Various polymers like HPMC, PVA, sodium alginate, and carbopol are utilized along with plasticizers and other excipients to achieve optimal film properties. Evaluation parameters such as thickness, folding endurance, surface pH, swelling index, drug content uniformity, in vitro disintegration, and drug release are discussed. This delivery system enhances bioavailability, ensures rapid onset of action, and improves patient compliance, making it a promising alternative to conventional dosage forms.
